Abstract:Abstract: Objective To breed Scamp2-/-/ApoE-/- mice and detect their serum high density lipoprotein cholesterol (HDL-C) and hepatic ABCA levels. Methods Scamp2-/- and ApoE-/- mice were raised in SPF room, and Scamp2-/-/ApoE-/- mice were bred using F0 homozygous hybridization and F1 heterozygous and F2 homozygous self-inbreeding. Genotypes of offspring were identified by PCR amplification and agarose gel electrophoresis. Serum HDL-C and hepatic ABCA1 mRNA levels were determined. Results Scamp2-/-/ApoE-/-, Scamp2-/-/ApoE+/+ and Scamp2+/+/ApoE-/- mice were successfully bred. Serum HDL-C and hepatic ABCA1 mRNA levels were lower in Scamp2-/-, Scamp2-/- and Scamp2-/-/ApoE-/- mice than in WT mice (P<0.001). Compared with ApoE-/- mice, serum HDL-C and hepatic ABCA1 mRNA levels were respectively decreased by 25.26% and 21.48% in Scamp2-/-/ApoE-/- mice (P<0.01). Conclusion Scamp2-/-/ApoE-/- mice are successfully obtained. Scamp2 knockout can reduce hepatic ABCA1 and serum HDL-C contents in mice.
