不同成分的饲料对葡聚糖硫酸钠诱导溃疡性结肠炎小鼠模型的影响

摘要:
目的 探讨不同成分的饲料对葡聚糖硫酸钠(DSS)诱导溃疡性结肠炎(UC)小鼠模型的影响。方法 68 只小鼠随机分为 2 组,分别饲喂饲料A(n=42)和饲料B(n=26),适应性喂养后随机分为对照组[ACtr组(n=21)和 BCtr组 (n=6)]和DSS组[A25 DSS组(n=21)、B25 DSS组(n=6)、B30 DSS组(n=6)、B35 DSS组(n=8)]。各组小鼠分别饲喂相应的饲料 11 d。对照组自由饮用无菌水 11 d。DSS组先自由饮用相应质量浓度DSS溶液 7 d,后改成饮用无菌水 4 d,实验结束后 评估小鼠身体质量、疾病活动度、结肠炎症状和死亡率等。结果 造模结束后,A25 DSS组小鼠的身体质量与B25 DSS、B30 DSS 和B35 DSS组差异无统计学意义(P>0.05)。A25 DSS组的疾病活动指数显著高于B25 DSS组(P<0.0001),与B30 DSS组和B35 DSS组 差异无统计学意义(P>0.05)。A25 DSS组小鼠的死亡率也显著高于B25 DSS、B30 DSS和B35 DSS组(P<0.001)。结论 营养比例 高的饲料可能有利于DSS诱导的UC模型的建立。

Effect of different feeds on dextran sodium-induced ulcerative colitis in mice

Abstract:
Objective To investigate the impact of different feeds on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice. Methods Sixty-eight mice were randomly allocated into two groups, each receiving feeds A (n=42) and B (n=26). After adaptive feeding, they were randomly divided into control group [ACtr group (n=21) and BCtr group (n=6)] and DSS group [A25 DSS group (n=21), B25 DSS group (n=6), B30 DSS group (n=6), B35 DSS group (n=8)]. Control group drank sterile water freely for 11 days. DSS group drank the corresponding concentration of DSS solution freely for 7 days, and then sterile water for 4 days. At the end of the experiment, the body mass, disease activity index (DAI), colitis symptoms and mortality were evaluated. Results After modeling, there was no significant difference in body weight among A25 DSS, B25 DSS, B30 DSS and B35 DSS groups (P>0.05). DAI of A25 DSS group was significantly higher than that of B25 DSS group (P<0.0001), but similar to B30 DSS and B35 DSS groups (P>0.05). The mortality was significantly higher in A25 DSS group than in B25 DSS, B30 DSS and B35 DSS groups (P<0.001). Conclusion High nutrient diet may be beneficial to the establishment of DSS-induced UC model.
